Summary: | Standard electroporation of Salmonella typhimurium TA1538 by the plasmid pAMH70 yielded 1.3x10 super(2) transformants/ mu g of plasmid DNA. Three parameters: resistance (U sub(1)), voltage (U sub(2)) and dose of plasmid DNA (U sub(3)) were optimized with two Doehlert designs. Transformation was increased 75 fold. Optimal values for U sub(1), U sub(2) and U sub(3) were 761.0 Omega , 1.6 kV and 96.1 ng respectively. The most important parameters were U sub(2) and the couple U sub(1)/U sub(2). |
