Summary: | Enzymes and their combinations were tested along with different osmotic stabilisers to produce protoplasts from mycelium of Botryotinia fuckeliana. Observations on cell wall regeneration and reversion to mycelial state were carried out on protoplasts plated on different media. Novozym 234, lysing enzyme and lyticase, especially in combinations including cellulase or ß-glucoronidase, resulted the most effective enzymes in KCl. Maximum release of protoplasts was achieved after 2-3 hours of treatment. Magnesium sulphate enhanced the effectiveness of novozym 234 + cellulase but reduced that of lyticase + ß-glucoronidase + cellulase + driselase. Proportion of colony-forming protoplasts (20-30%, generally) was similar on minimal or complete media but was influenced by protoplast origin, plating method and osmotic stabiliser contained in the medium. |
