Summary: | Xanthomonas perforans strain 91-118 produces at least three different bacteriocin-like compounds (BCN-A, BCN-B, BCN-C) of which two have been shown to give the strain a competitive advantage over Xanthomonas euvesicatoria on tomato in the field. Prior greenhouse and field experiments demonstrated that the bacteriocins conferred different levels of inhibition toward a X. euvesicatoria strain. Due to the pathogenicity of X. perforans, mutants were created to make X. perforans less pathogenic. An X. perforans mutant was created that was both attenuated in pathogenicity (opgH super(-)) and disrupted in BCN-B (Mut-B) activity to create the attenuated mutant Mut-B:DeltaopgH. In greenhouse growth rate and antagonism experiments internal populations of Mut-B:DeltaopgH was 1 to 1.5 log lower compared to the wild-type while maintaining its antagonism toward the X. euvesicatoria strain 91-106. Two field experiments in 2005 have also shown the ability of Mut-B:DeltaopgH to suppress X. euvesicatoria in the field. Further attenuated mutants are being evaluated for their ability to reduce X. euvesicatoria populations in planta while maintaining the ability to produce bacteriocins. |
