Summary: | Nucleoprotein complexes were isolated from blood plasma by affine chromatography using immobilized polyclonal antihistone antibodies. In blood of healthy subjects the prevalence of histone-containing nucleoprotein complexes, including 170–180 bp DNA, were found, whereas in blood of breast cancer patients the 170–180 bp DNA and the DNA containing more than 6000 bp were present in equal quantities. Using MALDI-TOF mass spectrometry, the proteins from histone-containing nucleoprotein complexes were identified that provided transport of extracellular nucleic acids into blood stream. It was found that nucleoprotein complexes from blood of breast cancer patients contained tumor-associated proteins, such as LDOC1L, ADP/ATP translocase 3, and Lamellipodin. These data confirm generation of part of circulating extracellular DNA by tumor cells. |
