Summary: | AFLPs have been used to characterize genetic diversity in 332 spinach genotypes in the USDA spinach germplasm collection. A total of 64 AFLP primer combinations were screened for their ability to identify polymorphisms among six spinach genotypes initially selected from diverse genetic pedigrees. The AFLP procedure yielded 16-124 reliably scorable fragments per primer combination. Several primer combinations, which yielded a high number of polymorphic fragments, were selected to evaluate the accessions in the germplams collection. The AFLP technique was a robust method for characterizing genetic diversity of spinach. The AFLP procedure was also used to identify polymorphisms among progeny of a cross between a downy mildew (Peronospora farinosa f. sp. spinaciae) susceptible and resistant parent (BC3). A single AFLP marker was found to be linked to the gene(s) controlling resistance to race 5 and 6 of the pathogen. |
