Summary:Highlights • Testicular macrophages (TM) comprise the largest leukocyte population in the testis and are M2 polarized. • TM bridge the tasks of maintaining immune privilege and responsiveness to infection by downstream blockade of TLR-4 signaling. • Instead moderate innate immune responses are mediated via MAP kinases and AP-1 and CREB transcription factors. • TM play also important roles in fetal vasculature development, Leydig cell steroidogenesis and spermatogonial stem cell function.
