Summary: | In 2001, a disease of melon characterized by yellowing of the leaves was observed in Sardinia (Italy), where it caused severe damage. Samples, collected from several melon crops, were positive in single step RT-PCR assays for Beet pseudoyellows virus (BPYV) when specific primers were used. An amplicon of the same size as that from BPYV controls (450 bp) was obtained from 14 out of 20 samples tested and had 98.4% nucleotide sequence identity with an authentic BPYV sequence. This is the first experimental evidence of a yellowing disease caused by BPYV in Italian melon crops
[Nel 2001, in Sardegna, e' stata osservata una malattia del melone, caratterizzata da ingiallimento delle foglie, che causava gravi danni. Campioni, raccolti da numerose colture di melone, sono risultati positivi a saggi RT-PCR a unico passaggio per il Virus del pseudoingiallimento della barbabietola (BPYV) con l'utilizzazione di primer specifici. E' stato ottenuto un amplicone della stessa dimensione di quello derivante dai controlli di BPYV (450 bp) da 14 dei 20 campioni provati e presentava un'identita' della sequenza nucleotidica pari al 98,4% rispetto a una sequenza autentica di BPYV.
