Description
Summary:The objective of the experiment was to design an efficient protocol for the production of gynogenic rubber tree (Hevea brasiliensis) embryos. Longitudinally bisected ovaries were cultured in five induction media. These media consisted of a modified Murashige and Skoog medium (medium called MBm) or Driver and Kuniyuki medium (DKW). The carbohydrates treatment consisted of 3.8% (111 mM) or 8% (234 mM) of sucrose and 2% (111 mM) of glucose. The growth regulator treatment was 9 mu M 2,4-D/2.25 mu M Kin or 9 mu M 2, 4-D/3.375 mu M Kin. Optimum embryos production was observed when induction medium was a modified mineral basic medium MB supplemented with 9 mu M 2, 4-D and 2.25 or 3.375 mu M Kin and 234 mM sucrose. Clones GT1, PB 217, PB 260 and PB 280 were studied. The embryogenic clones were PB 280, PB 260 and PB 217.
