Summary:To obtain sufficient material from isolated cells, a technique was used that amplified antisense RNA in a near linear manner. The labeled aRNA or its cDNA was hybridized to either reverse northern filters (up to 50 genes) or microarrays (>12000 genes). The profiles obtained from endothelium subjected to steady laminar flow in vitro and in cells isolated from carotid arteries showed consistent patterns of expression but variable magnitudes between cells. In contrast, transcription profiles were highly variable in both pattern and magnitude in regions of experimental flow disturbance.
