Here we reviewed the characteristics of the relationship between the postinjury immune response and heart regenerative capacity and summarized the latest studies focusing on inflammation and heart regeneration to identify potent targets of the immune response and strategies in the immune response to promote cardiac regeneration. •Acute immune response is mandatory for the initiation of cardiac regeneration in neonates.•Transplantation of pro-regenerative immune cells improves the outcome of cardiac repair.•IL-6 family cytokines actively regulate myocardium repair and are potent targets for cardiac regeneration.
