Summary:In this paper, the development of a hybrid polydimethylsiloxane (PDMS)-glass microchip for PCR (polymerase chain reaction) was reported. A novel temperature control strategy was applied for precise temperature control. The microchip was composed of two parts-a PDMS substrate and a glass cover. The PDMS substrate includes a reaction chamber for PCR and two microchannels for the inlet and the outlet of the sample. A glass cover integrating heaters and sensors was bonded to the PDMS substrate. The microchip was produced by molding PDMS against a microfabricated master with comparatively simple and inexpensive methods. PCR was successfully carried out on the PDMS-glass hybrid microchip with a 183 bp target of DNA.
