Summary: | Pre-Treatment of Mesenchymal Stem Cells With a Combination of Growth Factors Enhances Gap Junction Formation, Cytoprotective Effect on Cardiomyocytes, and Therapeutic Efficacy for Myocardial Infarction Joo-Yong Hahn, Hyun-Ju Cho, Hyun-Jae Kang, Tack-Seung Kim, Mi-Hyung Kim, Jung-Hwa Chung, Jang-Whan Bae, Byung-Hee Oh, Young-Bae Park, Hyo-Soo Kim Priming of mesenchymal stem cells (MSCs) with combination of fibroblast growth factor (GF)-2, insulin-like GF-1, and bone morphogenetic protein-2 enhanced the expression of gap junction and potentiated their cytoprotective effects on neighboring cardiomyocytes (CMCs). The cytoprotective effect on CMCs was mediated through gap junctions, which is a novel mechanism to explain therapeutic effect of MSC transplantation. Implantation of GF-treated MSCs after myocardial infarction reduced infarct size and improved left ventricular function. Priming of MSCs with cocktails of GFs might have important implication to enhance therapeutic efficacy for damaged myocardium. |
