Summary:Various kinds of polysaccharides (PSs) have been found to have antitumor activities and some of them are now used for the treatment of gastric cancer and uterine cancer. Schizophyllan is a PS produced extracellularly by Schizophyllum commune Fries. We have already succeeded in preparing a rabbit anti-schizophyllan antibody by immunizing rabbits subcutaneously with schizophylolan five times at two-week intervals. To further explore the nature of schizophyllan, we also decided to prepare a specific murine MAb, since there are few reports on the preparation of MAbs against PS by active immunization due to its low immunogenicity and lack of availability in pure form. Although there are several MAbs reactive with dextran, they were discovered only incidentally by immunologists while studying the antigen specificities of the MAbs. Thereafter, Kabat et al. have produced a large number of monoclonal anti-dextran antibodies by immunization for immunochemical studies. However, it is generally difficult to prepare MAbs to purified PS except dextran by active immunization.
