Summary:Using the RNase protection assay (RPA) to study the distribution of isoforms of the non-catalytic (β) subunit of Na,K-ATPase in the peripheral nervous system, we found both β1 and β2 isoform mRNAs in dorsal root ganglion (DRG), but only β2 mRNA in sciatic nerve. Using Western blot to measure accumulation of the polypeptides at a ligature on the nerve we found that β1 but not β2 polypeptide is carried by rapid axonal transport in the sciatic nerve. These results imply that β1 is the prominent isoform of Na,K-ATPase in neurons and β2 the prominent isoform in Schwann cells.
