Summary:The authors previously reported the construction of a DNA probe that recognizes DNA coding for production of Vi capsular antigen. The present study was undertaken to determine the sensitivity and specificity of this DNA probe, using in situ colony hybridization studies, to detect S. typhi among gram-negative bacteria freshly isolated from patients from two geographically distant areas. Initial studies conducted in Lima, Peru, were used to establish the conditions used in the subsequent study in Jakarta, Indonesia.
