Summary:The many advantages of Escherichia coli have ensured that it remains a valuable host for the efficient, cost-effective and high-level production of heterologous proteins. Here, we describe the current status of this prokaryotic expression system and focus on strategies designed to maximize the yields of recombinant proteins. Major challenges facing this expression system are also outlined.
