Summary:The insulin-like growth factors (IGFs) play important roles in regulating growth and development in vertebrates. Most of their actions are mediated by the IGF type 1 receptor (IGF-1R) which is a tetrameric protein composed of two alpha and beta subunits linked by disulfide bonds. The alpha and beta subunits arise from the processing of a single-chain preproreceptor. The alpha subunits, which are entirely extracellular, contain the ligand binding sites, whereas the beta subunits span the membrane and exhibit a tyrosine kinase domain in their cytoplasmic region. Cloning and sequencing of the complete IGF-1R cDNA have so far been reported only in two mammals, human and rat. In submammals, two cDNAs corresponding to a portion of the alpha and beta subunits have been described in chicken, whereas in Xenopus the sequence of a cDNA encoding nearly the entire beta subunit has been reported.
