Summary: | 1. Aspartate aminotransferase [EC 2.6.1.1] isoenzymes from supernatant and mito-chondrial fractions of dog heart muscle were crystallized and specific antibodies were prepared against each isoenzyme. 2. The purity of the enzyme was based upon the constant specific activity of the crystalline preparation, the presence of a single symmetrical peak in the analytical centrifugation pattern and the formation of a single immunoelectrophoretic arc. 3. The supernatant and mitochondrial isoenzymes had different Km values, electro-phoretic mobility, sedimentation coefficient, and immunological properties. 4. The immunological specificity of these enzymes seems to depend on the subcel-lular fraction used to prepare them and independent of the animal species and organ. |
