Summary: | Efficient colony formation of a purified line of continuously cultivable epithelial cells of the cottontail rabbit depended on the inclusion of both human adult and bovine fetal serum in medium. Human adult serum favored larger flattened cells, while only small, round cells formed colonies in bovine fetal serum alone. Mixed serum medium yielded many more colonies of each type. Serial isolation and dispersion of morphologically representative colonies and propagation of colonies from such dispersed cells showed that neither serum was adequate for continued cultivation of selected sublines. Cells serially propagated in human serum medium could not generate healthy colonies on return to mixed serum medium; cells serially propagated in bovine fetal serum medium lost colony-forming capacity but recovered it on return to mixed serum medium. |
