Summary:Several synthetic routes to the antimalarial agent 4-(2-bromo-4, 5-dimethoxyphenyl)-1,1,7, 7-tetraethyldiethylenetriamine (RC-12) (WR-27,653) were evaluated. The preferred scheme involved alkylation of 4-aminoveratrole with excess 2-chlorotriethylamine in dimethylformamide at 140–145° to give 4-(4, 5-dimethoxyphenyl)-1,1,7, 7-tetraethyldiethylenetriamine, which was brominated in acetic acid to give the desired product. The physical and biological properties of the drug are summarized.
