Summary:Collagen purification by ultrafiltration [1] is becoming widely accepted as a superior 14C sample preparation method for removing environmental contamination from archaeological bone. We present an overview of bone sample preparation methods used at the KCCAMS laboratory, UC Irvine. Methods development data are also discussed, including results of tests of gelatinization temperature, evaluation of 10 kDa vs. 30 kDa molecular weight (MW) ultrafiltration, improvements to ultrafilter cleaning methods, tests of ultrafilter memory, and ultrafilter glycerin contamination. These tests were not intended to be exhaustive, but rather to show how standard procedures can be validated and improved with relatively little effort.
