Summary:The isolation of a T. cruzi lambda gt11 recombinant cDNA clone, JL5, which reacted predominantly with chronic Chagas heart disease sera has been reported. The nucleotide and deduced amino acid sequence allowed identification as the C-terminal portion of a T. cruzi ribosomal P protein. Radioactive JL5 cDNA probes were used to screen a T. cruzi Tul2 stock epimastigote cDNA library. A recombinant was isolated, P-JL5, that encodes the complete amino acid sequence of the JL5 P protein. The underlined 17 bases at the 5' end represent the 3' region of the mini-exon sequence. The ORF of 107 amino acids begins at position 30. The first 7 N-terminal residues are 100% homologous to the yeast A2 P protein. The last 10 C-terminal residues define the distinctive consensus P protein motif. The serine at position 84 differs from the glycine reported for the JL5 sequence, due to the nucleotide change A(P-JL5) to G(JL5).
