Summary:The use of shuttle vectors to study DNA replication and repair in mammalian cells has created the need for techniques to recover plasmid DNA from mammalian cells. We describe a procedure that is rapid (< 2 h for 12 samples), simple (requires no phenol extractions), and provides DNA that is of a quality suitable for use directly in bacterial transformations or sequencing.
