Summary:We report the complete primary structure of rat liver mitochondrial prehsp60 deduced from cloned cDNA sequences. A lambda gt10 library prepared from adult rat liver mRNA was screened with oligonucleotides based on a human prehsp60 sequence and hybridizing cDNAs were sequenced. The entire coding region spans 1722 bases yielding a 573 amino acid primary translation product (M sub(r) 60,941) including a typical leader sequence of 26 residues. The predicted M sub(r) of the processed monomer is 57,912 in agreement with immunoblots of rat liver mitochondria using anti-hsp60 antibodies. The deduced amino acid sequence differs from that of human and Chinese hamster prehsp60 in 15 and 7 positions, respectively. (DBO)
