Summary:The bacterial blight pathogen of bean Xanthomonas axonopodis pv. phaseoli (hereinafter referred to as X. a. pv. phaseoli) affects many Phaseolus species. The increasing volume of legume yield in the Russian Federation and worldwide requires sensitive, reliable, and rapid methods to approve the absence of bacteriosis pathogen. These methods are based on conventional PCR tests. The objective of this work is to study, and to improve well known methods of diagnostics of bacterial blight pathogen of bean X. a. pv. phaseoli to be used with the commercial DNA extraction kits officially accepted in the Russian Federation. These commercial kits for DNA extraction and PCR methods show their effectiveness in the improved detection and identification assays of phytopathogen. After the full validation tests the adopted methods can be used as one of the official assays to detect X. a. pv. phaseoli in both symptomatic and asymptomatic plant and seed material.
