Summary:IntroductionAging-induced increase in fat accumulation causes arterial stiffness. In our recent study, local fat accumulation with aging may be more important than whole-body fat content for predicting cardiovascular disease. Aerobic exercise training (AT) decreases fat accumulation, concomitant with an elevation of serum level of adiponectin, as an anti-inflammatory adipokine, resulting reduction of arterial stiffness. Recently, C1q/TNF-related protein 3 (CTRP3) has been identified as a novel adipokine which is paralog of adiponectin and exerts vasodilator effect. Although adiponectin mRNA expression levels differ in various local fats, the differences of AT-induced increases in adiponectin and CTRP3 secretion from local adipose tissues remain unclear.HypothesisAT-induced elevations of adiponectin and CTRP3 secretion are associated with reduction of arterial stiffness in elderly, and these adipokines secretory responses differ between local fat cells.MethodsHuman study; 52 elderly subjects were randomly divided into AT and control (CON) groups. Subjects in the AT group completed 8-week of aerobic training (60-70% peak oxygen uptake for 45 min, 3 days/week). Animal study; seventeen 38-week-old SAMP1 mice were divided into CON and AT (voluntary wheel running for 12-weeks) groups. Expressions of adiponectin and CTRP3 mRNA in epididymal, subcutaneous and perivascular white fats and subscapular brown fat were measured by using real-time RT-PCR.ResultsIn human study, AT intervention significantly decreased cfPWV, as an index of arterial stiffness, and elevated serum adiponectin and CTRP3 levels (each P
